Nitrate-Induced MtCLE34 Gene Lacks the Ability to Reduce Symbiotic Nodule Number and Carries Nonsense Mutation in a Few Accessions of Medicago truncatula

چکیده

Legume plants form nitrogen-fixing nodules on their roots in symbiosis with soil bacteria rhizobia. The number of symbiotic is controlled by a host plant via systemic mechanism known as autoregulation nodulation (AON). key players AON are the CLE peptides which produced root response to rhizobia inoculation and transported xylem shoot. In shoot, recognized CLV1-like receptor kinase, results subsequent inhibition nodule development negative feedback mechanism. addition induced rhizobia, nitrate-induced involved control have been identified. Medicago truncatula, MtCLE34 gene has described, was activated nitrate rhizobial inoculation. However, this contains premature stop codon reference M. truncatula genome A17 line, therefore, it suggested be pseudogene. Here, we analyzed nucleotide sequences available from genomes different accessions HAPMAP project found that majority accession lines do not carry nonsense mutations should encode functional products. Overexpression R108 does codon, did inhibit nodulation. Therefore, spite having high sequence similarity nodulation-suppressing genes, line able trigger truncatula. Our findings shed light evolutionary changes proteins legume can used future understand amino acid residues within could important for ability suppress

Dual genetic pathways controlling nodule number in Medicago truncatula.

We report the isolation and characterization of a new Medicago truncatula hyper-nodulation mutant, designated sunn (super numeric nodules). Similar to the previously described ethylene-insensitive mutant sickle, sunn exhibits a 10-fold increase in the number of nodules within the primary nodulation zone. Despite this general similarity, these two mutants are readily distinguished based on anato...

A Medicago truncatula NADPH oxidase is involved in symbiotic nodule functioning

The plant plasma membrane-localized NADPH oxidases, known as respiratory burst oxidase homologues (RBOHs), appear to play crucial roles in plant growth and development. They are involved in important processes, such as root hair growth, plant defence reactions and abscisic acid signalling. Using sequence similarity searches, we identified seven putative RBOH-encoding genes in the Medicago trunc...

MAP Kinase-Mediated Negative Regulation of Symbiotic Nodule Formation in Medicago truncatula

Mitogen-activated protein kinase (MAPK) signaling cascades play critical roles in various cellular events in plants, including stress responses, innate immunity, hormone signaling, and cell specificity. MAPK-mediated stress signaling is also known to negatively regulate nitrogen-fixing symbiotic interactions, but the molecular mechanism of the MAPK signaling cascades underlying the symbiotic no...

A CDPK isoform participates in the regulation of nodule number in Medicago truncatula.

Medicago spp. are able to develop root nodules via symbiotic interaction with Sinorhizobium meliloti. Calcium-dependent protein kinases (CDPKs) are involved in various signalling pathways in plants, and we found that expression of MtCPK3, a CDPK isoform present in roots of the model legume Medicago truncatula, is regulated during the nodulation process. Early inductions were detected 15 min and...
